High-Sensitivity Proximity Assays
Ideally, biological assays will yield quick, clear results through sensitive and selective detection in complex mixtures. High-sensitivity proximity assays are favored for facilitating high-throughput screening and revealing relative distances between analytes. To expand the utility of a proximity assay, SRI researchers have dramatically increased their sensitivity. The technology is available for license.
Assays that use luminescent lanthanide complexes for signaling have the advantages of providing sharp, discrete peaks and long emissions. However, they lack brightness, which hampers overall sensitivity. Meanwhile, traditional organic fluors have short shelf lives and broad peaks that limit potential applications and preclude use for revealing more than one analyte at a time.
SRI’s approach combines lanthanide labeling with surface-enhanced fluorescence. The result is a high-sensitivity proximity assay that has greater sensitivity and that retains sharp emission peaks, which are easily discriminated from excitation wavelengths.
